Argyle have been drawn away to fellow Sky Bet League One side Leyton Orient in the second round of the Vertu Trophy.
The Greens finished second in their group, having beaten Cheltenham Town and Tottenham Hotspur Under-21s, but then lost to Bristol Rovers, finishing behind the Gas in the Group B table.
Orient won Group F, winning all three of their group stages games, against Peterborough United, Crawley Town, and Aston Villa Under-21s.
Orient, who are managed by Richie Wellens, are currently 20th in the League One table, and won 1-0 at Home Park earlier in the season.
Argyle have played Orient four times in the last dozen years in the Carabao Cup, but the sides have never met in this Trophy.
The tie is set to take place in the week commencing 1 December. An exact date and ticket details will be announced when agreed.
